New records and hosts of the Curvularia species in Iran

Abstract

In present study, eight isolates of the genus Curvularia, recovered from plants of the cowpea, maize and bottlebrush in Khuzestan province, were identified based on molecular phylogeny in combination with morphology. The mycelial biomass of the isolates, produced in potato-dextrose-Broth, were collected by passing through filter papers and freeze-dried. DNA was extracted using a phenol- and chloroform-based method. The partial regions of nrDNA and gpd were amplified using the appropriate primers and then sequenced. The consensus ITS and gpd sequences of the isolates under survey were compared to reference sequences of the type strains in GenBank by BLASTn algorithm and two-loci phylogenetic analysis. Accordingly, the isolates were identified at species level as follow: Curvularia americana, C. muehlenbeckiae,and C.verruculosa. To our knowledge, this is the first record of both the C. americana and C. muehlenbeckiae in Iran. Moreover, this study reports new hosts for all three species with further morphological and molecular characterization of C.verruculosa species as well.

Abstract [Persian]

در مطالعه حاضر، هشت جدایه‏‌‌‌‌‌ از جنس Curvularia، جداسازی شده از گیاهان لوبیا چشم‌بلبلی، ذرت و شیشه‌شور در استان خوزستان، براساس روش‏‌‌‌‌‌ مبتنی بر فیلوژنی مولکولی و ریخت‏‌‌‌‌‌شناسی شناسایی شدند. زیست‏‌‌‌‌‌توده‌ میسلیومی جدایه‏‌‌‌‌‌ها، تولید شده در محیط مایع سیب زمینی-دکستروز، به وسیله‏‌‌‌‌‌ کاغذ صافی جمع‏‌‌‌‌‌آوری و خشک-‏‌‌‌‌‌انجمادی شد. توده‏‌‌‌‌‌ DNA با استفاده از یک روش مبتنی بر فنل و کلروفرم استخراج شد. بخش‏‌‌‌‌‌هایی از نواحی rDNA هسته‏‌‌‌‌‌ای و gpd با استفاده از آغازگر‏‌‌‌‌‌های مناسب تکثیر و سپس توالی‏‌‌‌‌‌یابی شدند. توالی‏‌‌‌‌‌های ITS و gpd مربوط به جدایه‏‌‌‌‌‌های تحت تحقیق با توالی‏‌‌‌‌‌های معتبر مربوط به سویه‏‌‌‌‌‌های تیپ در بانک ژن با استفاده از الگوریتم BLASTn و تجزیه و تحلیل فیلوژنتیکی (مبتنی بر دو ناحیه‏‌‌‌‌‌ ژنومی) مورد مقایسه قرار گرفتند. بر این اساس، جدایه‏‌‌‌‌‌ها در سطح گونه شناسایی و گونه‏‌‌‌‌‌هایC. americana، C. muehlenbeckiae و C.verruculosa را شامل شدند. لذا، این نخستین گزارش دو گونه‏‌‌‌‌‌ C. americana و C. muehlenbeckiae در ایران می‏‌‌‌‌‌باشد. به علاوه، این مطالعه برای هر سه گونه‌‌‌‌‌ مورد بررسی میزبان جدیدی گزارش می‏‌‌‌‌‌کند و مشخصات ریخت‏‌‌‌شناسی و مولکولی بیشتری برای گونه‌ C.verruculosa ارایه می‏‌‌‌‌‌دهد.
